Niclosamide is an FDA-approved drug that can elicit anti-tumor effects via direct STAT3 inhibition, which has been revealed in numerous human cancer cells. Chemotherapy is the standard treatment for advanced esophageal cancer but causes severe systemic side effects. The present study represents the first study to evaluate the anticancer efficacy of niclosamide in esophageal cancer. By Western blot analysis, it has been demonstrated that niclosamide inhibits STAT3 signaling pathways in esophageal adenocarcinoma cells (BE3) and esophageal squamous cell carcinoma cells (CE48T and CE81T).

Furthermore, niclosamide is an oral antifungal agent used to treat parasitic infections in millions of people around the world. However, according to recent studies, niclosamide may have wide clinical applications for the treatment of diseases other than those caused by parasites. These diseases and symptoms may include cancer, bacterial and viral infections, type II diabetes, metabolic diseases such as NASH and NAFLD, arterial stenosis, endometriosis, neuropathic pain, rheumatoid arthritis, sclerotic graft-to-host disease, and systemic sclerosis. Among the basic mechanisms involved in the drug action of niclosamide are the isolation of oxidative phosphorylation and the regulation of Wnt/β-catenin, mTORC1, STAT3, NF-κB, and notch signaling pathways.

Usage/capacity

No special preparation or additional steps (e.g., special diet, fasting, other medications, laxatives, or enemas) are required before, during, or immediately after taking closamide. Niclosamide can be taken on an empty stomach (one or two hours after meals).

However, it is best to take it after a light meal (e.g., breakfast) to prevent gastrointestinal disorders.

Niclosamide tablets should be thoroughly chewed or ground and then swallowed with a small amount of water.

When this medicine is given to a young child, the tablet should be ground into fine powder and mixed with a small amount of water to make a paste.

For patients taking beef tapeworm or broad or tapeworm A drugs:

To completely eliminate the infection, follow your doctor’s instructions to take this medicine. Usually, one time is enough.

However, for some patients, a second dose of this drug may be necessary to completely eliminate the infection.

For patients taking this medication against Dwarf tapeworm:

To completely eliminate the infection, continue taking this medication (usually 7 days) for the duration of your treatment, even if the symptoms begin to disappear after a few days.

Some patients may need a second course of this drug to completely eliminate the infection.

If you stop taking this medicine too soon, the infection may reoccur.

For fish tapeworms or beef tapeworms:

Adult – 2g in a single dose. Treatment can be repeated in 7 days if needed.

Child-dose is based on body weight and should be determined by the doctor. Children weighing 11 to 34 kg (24.2 to 74.8 pounds) weigh 1 g at a time. Treatment can be repeated within 7 days if necessary. For children weighing over 34 kg (74.8 pounds), 1.5 g in a single dose. Treatment can be repeated within 7 days if necessary.

For dwarf tapeworms:

Adult —2 g per day for 7 days. Treatment can be repeated after 7 to 14 days if required.

Child-dose is based on weight and should be determined by the doctor. Children: 11 to 34 kg (24.2 to 74.8 pounds) 1 g on day one: then 500 milligrams (mg) once a day for the next 6 days. Treatment can be repeated after 7 to 14 days if needed. For children who weigh more than 34 kg (74.8 pounds): 1.5 g on day one. Take 1 gram per day for the next 6 days. Treatment can be repeated after 7 to 14 days if needed.
